چکیده

The multi-step synthesis of complex organic compounds from simpler precursors is one of the outstanding accomplishments and ongoing challenges of synthetic organic chemistry. Through the development and invention of synthesis strategies, methods and technologies, increasingly complex molecules can be assembled with designed structures and functions for a variety of medicinal, agrochemical and materials applications. However, despite significant advances, organic synthesis is still considered highly labourand resource-intensive. The traditional pathway for multi-step synthesis proceeds by the batchwise and iterative step-by-step transformation of starting materials into desired products (Fig. 1(a)). Typically, after the completion of each synthetic step (A + B / C, C / D and D / E), products are isolated from the reaction mixture and purified to remove any undesired components that might interfere with the subsequent synthetic transformations. Although this approach is the foundation on which modern synthesis has been built, such an approach is time-consuming, often wasteful and in stark contrast to the single-cell multi-step biosynthetic pathways found in nature.

Semi-continuous multi-step synthesis of lamivudine.

We report the first continuous flow synthesis of lamivudine, an antiretroviral drug used in the treatment of HIV/AIDS and hepatitis B. The key intermediate (5-acetoxy oxathiolane) was prepared by an integrated two step continuous flow process from l-menthyl glyoxalate hydrate in a single solvent, in 95% overall conversion. For the crucial glycosidation reaction, using pyridinium triflate as the...

Accessing Stereochemically Rich Sultams via Microwave-Assisted, Continuous Flow Organic Synthesis (MACOS) Scale-out.

The generation of stereochemically-rich benzothiaoxazepine-1,1'-dioxides for enrichment of high-throughput screening collections is reported. Utilizing a microwave-assisted, continuous flow organic synthesis platform (MACOS), scale-out of core benzothiaoxazepine-1,1'-dioxide scaffolds has been achieved on multi-gram scale using an epoxide opening/S(N)Ar cyclization protocol. Diversification of ...

Continuous-flow synthesis of highly functionalized imidazo-oxadiazoles facilitated by microfluidic extraction

A versatile continuous-flow synthesis of highly functionalized 1,2,4-oxadiazoles starting from carboxylic acids is reported. This process was applied to the multistep synthesis of imidazo[1,2-a]pyridin-2-yl-1,2,4-oxadiazoles, using a three reactor, multistep continuous-flow system without isolation of intermediates. This continuous-flow method was successfully combined with a single-step liquid...

One-pot Multi-step Synthesis of some Aromatic Salicylaldoximes Using MgO Nanoparticles

MgO nanopartticels (NP-MgO) in average size between 30—130 nm were prepared by sonochemistry method. Some phenols have been converted to ortho-hydroxybezaldehyd by ortho-formylation using MgO nanoparticles and (CH2O)n in xylene and subsequently treated with aqueous NH2OH.HCl, affording the corresponding aldoxime in a one-pot procedure. Moreover, this one-pot process, with two transformations, o...

Continuous flow synthesis and scale-up of glycine- and taurine-conjugated bile salts.

A multi-gram scale protocol for the N-acyl amidation of bile acids with glycine and taurine has been successfully developed under continuous flow processing conditions. Selecting ursodeoxycholic acid (UDCA) as the model compound and N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline (EEDQ) as the condensing agent, a modular mesoreactor assisted flow set-up was employed to significantly speed up the...
